    NAVIGATING VIDEO-BASED LEARNING IN SCIENCE ā€“ HOW DO WE CLOSE THE GAP BETWEEN ONLINE AND PHYSICAL CLASSROOMS?

    Online learning increases the physical distance between instructors and students and depending on the mode of delivery it can be challenging to close this gap. In the face of a global pandemic institutions are rapidly increasing their portfolio of online and blended courses, which increases the potential risk of student attrition across the sector. To ameliorate this potential for student isolation, instructors need to communicate to students in a variety of ways, blending original online resources with synchronous interactive learning activities. In 2020, 55 videos designed for lectures, tutorials, and laboratory sessions were created for 400 undergraduate microbiology students at The University of Queensland. The videos collectively received over 35,000 views, and through learning analytics it was observed that videos featuring instructor presence on screen, frequent scene transitions, and dynamic pacing increased the consistency of student engagement (>80% average audience retention). This session will outline the design principles underlying a framework for developing video-based learning resources in science to maximise their utility as part of blended and online courses, as well as the academic digital upskilling required in the current Higher Education landscape

    Characterization of soluble bromide measurements and a case study of BrO observations during ARCTAS

    A focus of the Arctic Research of the Composition of the Troposphere from Aircraft and Satellites (ARCTAS) mission was examination of bromine photochemistry in the spring time high latitude troposphere based on aircraft and satellite measurements of bromine oxide (BrO) and related species. The NASA DC-8 aircraft utilized a chemical ionization mass spectrometer (CIMS) to measure BrO and a mist chamber (MC) to measure soluble bromide. We have determined that the MC detection efficiency to molecular bromine (Br2), hypobromous acid (HOBr), bromine oxide (BrO), and hydrogen bromide (HBr) as soluble bromide (Brāˆ’) was 0.9Ā±0.1, 1.06+0.30/āˆ’0.35, 0.4Ā±0.1, and 0.95Ā±0.1, respectively. These efficiency factors were used to estimate soluble bromide levels along the DC-8 flight track of 17 April 2008 from photochemical calculations constrained to in situ BrO measured by CIMS. During this flight, the highest levels of soluble bromide and BrO were observed and atmospheric conditions were ideal for the space-borne observation of BrO. The good agreement (R2 = 0.76; slope = 0.95; intercept = āˆ’3.4 pmol molāˆ’1) between modeled and observed soluble bromide, when BrO was above detection limit (\u3e2 pmol molāˆ’1) under unpolluted conditions (NOmolāˆ’1), indicates that the CIMS BrO measurements were consistent with the MC soluble bromide and that a well characterized MC can be used to derive mixing ratios of some reactive bromine compounds. Tropospheric BrO vertical column densities (BrOVCD) derived from CIMS BrO observations compare well with BrOTROPVCD from OMI on 17 April 2008

    Undergraduate Research Experiences: a Case Study in promoting Student Engagement through Authentic Scientific Inquiry

    Laboratory exercises teach practical skills that form the foundation of scientific research. These classes however, have been ineffective in promoting student engagement in science, as they are often structured around rigid and repetitive protocols. These traditional ā€œcookbookā€ laboratory classes are not an accurate representation of scientific inquiry, and do not teach students the autonomy required to succeed as a professional scientist. This project directly engaged students in the discovery process by integrating Undergraduate Research Experience (URE) modules into MICR3003, a third year Microbiology course offered at the University of Queensland (UQ). As part of their undergraduate coursework, students conducted inquiry-based experiments to make novel experimental findings, and were assessed on their adherence to professional scientific standards. At the end of the URE, students demonstrated improvements in key experimental, reporting, and analytical skills, as well as an increase in their general interest in science; moreover, the URE participants appreciated the opportunity to collectively experience an immersive undergraduate research project. Embedding active research projects into the undergraduate curriculum is able to reach a far greater number of students than isolated laboratory internships, and thus is an effective mechanism for increasing exposure for science and providing training for the next generation of scientists

    Combining Anomaly and Z' Mediation of Supersymmetry Breaking

    We propose a scenario in which the supersymmetry breaking effect mediated by an additional U(1)' is comparable with that of anomaly mediation. We argue that such a scenario can be naturally realized in a large class of models. Combining anomaly with Z' mediation allows us to solve the tachyonic slepton problem of the former and avoid significant fine tuning in the latter. We focus on an NMSSM-like scenario where U(1)' gauge invariance is used to forbid a tree-level mu term, and present concrete models, which admit successful dynamical electroweak symmetry breaking. Gaugino masses are somewhat lighter than the scalar masses, and the third generation squarks are lighter than the first two. In the specific class of models under consideration, the gluino is light since it only receives a contribution from 2-loop anomaly mediation, and it decays dominantly into third generation quarks. Gluino production leads to distinct LHC signals and prospects of early discovery. In addition, there is a relatively light Z', with mass in the range of several TeV.     DNA replication stress restricts ribosomal DNA copy number

    Ribosomal RNAs (rRNAs) in budding yeast are encoded by ~100ā€“200 repeats of a 9.1kb sequence arranged in tandem on chromosome XII, the ribosomal DNA (rDNA) locus. Copy number of rDNA repeat units in eukaryotic cells is maintained far in excess of the requirement for ribosome biogenesis. Despite the importance of the repeats for both ribosomal and non-ribosomal functions, it is currently not known how ā€œnormalā€ copy number is determined or maintained. To identify essential genes involved in the maintenance of rDNA copy number, we developed a droplet digital PCR based assay to measure rDNA copy number in yeast and used it to screen a yeast conditional temperature-sensitive mutant collection of essential genes. Our screen revealed that low rDNA copy number is associated with compromised DNA replication. Further, subculturing yeast under two separate conditions of DNA replication stress selected for a contraction of the rDNA array independent of the replication fork blocking protein, Fob1. Interestingly, cells with a contracted array grew better than their counterparts with normal copy number under conditions of DNA replication stress. Our data indicate that DNA replication stresses select for a smaller rDNA array. We speculate that this liberates scarce replication factors for use by the rest of the genome, which in turn helps cells complete DNA replication and continue to propagate. Interestingly, tumors from mini chromosome maintenance 2 (MCM2)-deficient mice also show a loss of rDNA repeats. Our data suggest that a reduction in rDNA copy number may indicate a history of DNA replication stress, and that rDNA array size could serve as a diagnostic marker for replication stress.     Realistic Standard Model Fermion Mass Relations in Generalized Minimal Supergravity (GmSUGRA)

    Grand Unified Theories (GUTs) usually predict wrong Standard Model (SM) fermion mass relation m_e/m_{\mu} = m_d/m_s toward low energies. To solve this problem, we consider the Generalized Minimal Supergravity (GmSUGRA) models, which are GUTs with gravity mediated supersymmetry breaking and higher dimensional operators. Introducing non-renormalizable terms in the super- and K\"ahler potentials, we can obtain the correct SM fermion mass relations in the SU(5) model with GUT Higgs fields in the {\bf 24} and {\bf 75} representations, and in the SO(10) model. In the latter case the gauge symmetry is broken down to SU(3)_C X SU(2)_L X SU(2)_R X U(1)_{B-L}, to flipped SU(5)X U(1)_X, or to SU(3)_C X SU(2)_L X U(1)_1 X U(1)_2.     BackgroundSpinal Toll-like receptors (TLRs) and signaling intermediaries have been implicated in persistent pain states. We examined the roles of two major TLR signaling pathways and selected TLRs in a mononeuropathic allodynia.MethodsL5 spinal nerve ligation (SNL) was performed in wild type (WT, C57BL/6) male and female mice and in male Tlr2-/-Tlr3-/-, Tlr4-/-, Tlr5-/-, Myd88-/-, Triflps2, Myd88/Triflps2, Tnf-/-, and Ifnar1-/- mice. We also examined L5 ligation in Tlr4-/- female mice. We examined tactile allodynia using von Frey hairs. Iba-1 (microglia) and GFAP (astrocytes) were assessed in spinal cords by immunostaining. Tactile thresholds were analyzed by 1- and 2-way ANOVA and the Bonferroni post hoc test was used.ResultsIn WT male and female mice, SNL lesions resulted in a persistent and robust ipsilateral, tactile allodynia. In males with TLR2, 3, 4, or 5 deficiencies, tactile allodynia was significantly, but incompletely, reversed (approximately 50%) as compared to WT. This effect was not seen in female Tlr4-/- mice. Increases in ipsilateral lumbar Iba-1 and GFAP were seen in mutant and WT mice. Mice deficient in MyD88, or MyD88 and TRIF, showed an approximately 50% reduction in withdrawal thresholds and reduced ipsilateral Iba-1. In contrast, TRIF and interferon receptor null mice developed a profound ipsilateral and contralateral tactile allodynia. In lumbar sections of the spinal cords, we observed a greater increase in Iba-1 immunoreactivity in the TRIF-signaling deficient mice as compared to WT, but no significant increase in GFAP. Removing MyD88 abrogated the contralateral allodynia in the TRIF signaling-deficient mice. Conversely, IFNĪ², released downstream to TRIF signaling, administered intrathecally, temporarily reversed the tactile allodynia.ConclusionsThese observations suggest a critical role for the MyD88 pathway in initiating neuropathic pain, but a distinct role for the TRIF pathway and interferon in regulating neuropathic pain phenotypes in male mice
